Measurement of Citrate Synthase Activity in a Microplate Format

Background: Citrate synthase (CS) activity is commonly used as a marker for mitochondrial intactness and content. The activity can also be used as a normalization factor for mitochondrial proteins and enzyme activities. Previous methods measured the CS activity using spectrophotometer in a cuvette that required a larger amount of reagents and sample input. This study aimed to measure the CS activity in a 96-well plate, a microplate format.
Methods: Mitochondria were isolated from rat hippocampus and the CS activity was measured using spectrophotometer in a 96-well plate.
